Comet assay comparison of different Corbicula fluminea (Mollusca) tissues for the detection of genotoxicity Genet. Mol. Biol.

The comet assay was used to study the sensitivity of the widely distributed freshwater bivalve mollusk Corbicula fluminea to the DNA-damaging alkylating-agent methylmethane sulfonate (MMS). This study was undertaken to ascertain if C. fluminea is a good bioindicator of pollutants in aquatic environments and identify which C. fluminea tissue is most effective and practical for genotoxicity studies. The mollusks were exposed to 0.6, 1.2 or 2.4 X 10-4 M MMS for 40 min and their hemolymph, gill tissue and digestive gland tissue assessed for the level of DNA damage and the time needed for the tissues to recovery. Cytotoxic and antibacterial properties of Mytilus galloprovincialis, Ostrea edulis and Crassostrea gigas (bivalve molluscs) hemolymph ArchiMer

Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45°C. Large individual variability which was not correlated with protein concentration and an increasing number of reactive specimens following injection, suggested that naive mussels were in various stages of immune response. The hemocyte composition of adults of Amblyomma cajennense (Fabricius, 1787) Koch, 1844 adults consisted of three basic cellular types: prahemocyte I (PR I), plasmatocyte (PL) and spherulocyte (ES II). Among the ixodids observed (A. cajennense; A. dissimile Koch, 1844; Rhipicephalus sanguineus Latreille, 1806; Anocentor nitens (Neumann, 1897) Schulze, 1937; Boophilus microplus Canestrini, 1887 and Haemaphysalis sp.) only A. cajennense was different in the adult hemolymph colour. It changed from amber to deep blue as the ixodid age advanced. Estudo morfológico e quantitativo dos hemócitos em larvas de Diatraea saccharalis (Fabricius) (Lepidoptera, Pyralidae) Zoologia

Hemocytes of Diatraea saccharalis larvae aging from 13 to 26 days old were morphological and quantitatively characterized by phase contrast microscopy. Six hemocyte types were described and identified as: prohemocytes (PR), plasmatocytes (PL), granulocytes (GR), oenocytoides (OE). spherulocytes (SP) and verm cells (VE). The total hemocyte counts showed no significant difference in the number of these cells along the normal development of the larvae. However, the relative number of different circulating hemocyte types fluctuated, and it could be expressed as: PL > GR > SP> PR > VE > OE in larvae up to 14 days old; and as: GR > SP > PL > VE > OE > PR in those older than 24 days.

Induction of a gloverin-like antimicrobial polypeptide in the sugarcane borer Diatraea saccharalis challenged by septic injury BJMBR

Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) is an important pest for Brazilian sugarcane. In the present study, we detected two distinct spots in hemolymph from septic injured larvae (HDs1 and HDs2), which are separated by 2DE gel electrophoresis. Both spots were subjected to in-gel tryptic digestion and MALDI-TOF/TOF analysis, which revealed the sequence VFGTLGSDDSGLFGK present in both HDs1 and HDs2. This sequence had homology and 80% identity with specific Lepidoptera antimicrobial peptides called gloverins. Analyses using the ImageMaster 2D software showed pI 8.94 of the HDs1 spot, which is similar to that described to Hyalophora gloveri gloverin (pI 8.5). Performance evaluation of CHO-K1 cell in culture medium supplemented with hemolymph BABT

The aim of this work was to evaluate the potential of hemolymph utilization as a culture medium supplement to cultivate the animal cell CHO-K1. For this purpose 1% v/v of hemolymph was added to DMEM medium containing 10% v/v of FBS and 1 or 4.5 g/L of glucose. The culture was grown in spinner flasks incubated in a 10% v/v CO2 environment, at 37ºC, with the Cytodex 1 microcarrier. Comparing the results obtained from the culture with hemolymph against those without hemolymph, a positive influence of the hemolymph was observed, as the experiment with hemolymph presented a 52% higher cell concentration and a higher productivity of up to 40%.

Little is known about possible antiviral substances in invertebrates. The present work concerns a Study of antiviral substances in hemolymph from adult Crassostrea gigas oysters. Worker honeybee hemolymph lipid composition and synodic lunar cycle periodicities BJMBR
The aim of the present investigation was to extend a previous study, showing a correlation of the variations of hemolymph carbohydrates with synodic lunar-like cycle and its circaseptan harmonics to worker honeybee hemolymph lipids. Hemolymph lipid concentrations of emerging worker imagos were analyzed in terms of one ideal synodic lunar cycle and processed by the cosinor method testing the null hypothesis versus the presence of 29.5-, 14.8- or 7.4-day periods in the data. A rhythmicity statistically compatible with a 29.5-day rhythm was observed for triacylglycerols and steroids as well as for body weight. Swelling and lysis of hemolymph erythroid elements of Anadara inaequivalvis were observed under experimental anoxia conditions. Mainly late generation cells was destroyed. It’s accompanied by increase of geometrical parameters of erythrocytes and their nucleuses.
